Recombinant Human T cell receptor delta variable 2 (TRDV2) fragment (amino acids 20-115) is a high-purity, soluble protein designed for structural and functional studies. This 96-amino acid fragment represents a stable variable domain region ideal for investigating gamma-delta T cell receptor biology, antigen recognition mechanisms, and immune signaling pathways.
The full-length T cell receptor delta variable 2 protein (UniProt ID: A0JD36) functions as the V region of the variable domain in T cell receptor delta chains, playing a critical role in antigen recognition. Gamma-delta T cell receptors recognize diverse self and foreign non-peptide antigens at epithelial boundaries, including endogenous lipids presented by CD1D and phosphoantigens presented by BTN3A1. Upon antigen binding, these receptors trigger rapid, innate-like immune responses essential for pathogen clearance and tissue repair. The signaling cascade involves phosphorylation of immunoreceptor tyrosine-based activation motifs (ITAMs) in CD3 chains by LCK and FYN kinases, leading to ZAP70 recruitment and activation. This initiates formation of a supramolecular signalosome that recruits PLCG1, resulting in calcium mobilization and ERK activation, ultimately driving T cell expansion and differentiation.
