Serine/arginine-rich splicing factor 2 (SRSF2/SC35) is a crucial nuclear protein essential for pre-mRNA splicing in human cells. It plays a fundamental role in the formation of the earliest ATP-dependent splicing complex and mediates interactions between spliceosomal components bound to both 5'- and 3'-splice sites during spliceosome assembly. The protein binds to purine-rich RNA sequences and is required for ATP-dependent interactions of U1 and U2 snRNPs with pre-mRNA. SRSF2 localizes primarily to nuclear speckles, though phosphorylation by SRPK2 triggers redistribution to the nucleoplasm and is involved in cellular apoptosis responses to cisplatin treatment. The full-length protein functions as a bridging component between 5'- and 3'-splice site binding elements through its RS domains.
