Intraflagellar transport protein 20 homolog (IFT20) is a critical component of intraflagellar transport particles essential for ciliary assembly and function in human cells. It facilitates the trafficking of ciliary membrane proteins from the Golgi complex to the cilium and regulates the platelet-derived growth factor receptor-alpha (PDGFRA) signaling pathway by maintaining the stability of E3 ubiquitin ligases CBL and CBLB. IFT20 is also vital for male fertility, playing key roles in spermatogenesis, flagella formation, and the transport of proteins like ODF2 and SPAG16. Additionally, it contributes to autophagy by enabling the trafficking of ATG16L and expansion of autophagic compartments. The protein localizes to multiple subcellular sites, including the Golgi apparatus, centrosomes, basal bodies, cilia, and cytoplasmic vesicles, underscoring its diverse functional roles in cellular processes.
