Description

IFIT1 is a key effector in the innate immune response, forming part of an interferon-dependent multiprotein complex that discriminates and sequesters viral RNAs lacking host-specific 2'-O-methylation at their 5' cap. By competing with the translation initiation factor eIF4E, it inhibits the translation of these viral RNAs, thereby restricting viral replication. Additionally, IFIT1 interacts with viral proteins, such as the DNA replication origin-binding protein E1 of certain viruses, causing their translocation from the nucleus to the cytoplasm and inhibiting helicase activity. It exhibits broad antiviral activity against viruses from families including Flaviviridae (e.g., West Nile, Dengue, and hepatitis C viruses), Coronaviridae (e.g., human 229E coronavirus, SARS-CoV-2, and SARS-CoV), Poxviridae (e.g., vaccinia virus), and Togaviridae (e.g., Sindbis virus). IFIT1 is localized in the cytoplasm, where it executes its antiviral functions.
