Dimethyladenosine transferase (DIMT1) is a crucial enzyme that specifically dimethylates two adjacent adenosines (A1779 and A1780) in the conserved hairpin loop near the 3'-end of 18S rRNA within the 40S ribosomal subunit. Beyond its catalytic RNA-modifying function, DIMT1 plays an essential role in pre-rRNA processing steps required for small-subunit rRNA production. The protein is a component of the small subunit (SSU) processome, the primary precursor of the small eukaryotic ribosomal subunit. During SSU processome assembly in the nucleolus, DIMT1 collaborates with numerous ribosome biogenesis factors, RNA chaperones, and ribosomal proteins to facilitate proper RNA folding, modifications, structural rearrangements, and cleavage events, as well as targeted degradation of pre-ribosomal RNA by the RNA exosome. The protein localizes to both the nucleoplasm and nucleolus, reflecting its dynamic role in nuclear ribosomal biogenesis processes.
