    CLE45 is a peptide related to CLAVATA3/EMBRYO SURROUNDING REIGON. It is genuinely expressed in developing protophloem sieve elements (PPSEs). Autocrine CLE45 signaling through BAM3 must be dampened below a threshold level to permit protophloem differentiation. It also acts paracrine to keep neighboring cell files in a pluripotent state through signaling via RPK2. The wild type becomes insensitive to CLE45 treatment on neutral to alkaline pH media, and pH-dependent perception specifically requires the CLE45 R4 residue and is not observed for CLE25 and CLE26. CLE45 is a potent inhibitor of protophloem formation and root growth. It has a different structure from CLE25 and CLE26, differing from CLE26 in positions 4 and 7, and from CLE25 in multiple positions. A synthetic mutant variant with certain residue replacements shows different behaviors related to pH dependence and receptor recognition.
