Apolipoprotein C-III (Apo-CIII) is a key component of triglyceride-rich very low density lipoproteins (VLDL) and high density lipoproteins (HDL) in human plasma. It plays a multifaceted role in triglyceride homeostasis by promoting hepatic VLDL1 assembly and secretion intracellularly, while extracellularly it attenuates the hydrolysis and clearance of triglyceride-rich lipoproteins. Apo-CIII impairs lipolysis by inhibiting lipoprotein lipase and hinders hepatic uptake of these lipoproteins by remnant receptors. Structurally, it consists of several curved helices connected via flexible hinges, allowing it to adapt to and wrap around micelle surfaces of varying diameters. The protein is secreted and functions primarily in plasma, making it a significant target for metabolic and cardiovascular research.
