Proline-rich AKT1 substrate 1 (AKT1S1) is a human protein that functions as a critical negative regulator of the mechanistic target of rapamycin complex 1 (mTORC1), an evolutionarily conserved nutrient sensor that promotes anabolic reactions and macromolecule biosynthesis to drive cellular growth. In the absence of insulin and nutrients, AKT1S1 associates with mTORC1 and directly inhibits its activity by blocking the substrate-recruitment site. Upon insulin or nutrient stimulation, AKT1S1 dissociates from mTORC1, with this regulatory mechanism being dependent on its phosphorylation state and interaction with 14-3-3 proteins. AKT1S1 is primarily localized in the cytoplasm and cytosol, and it may also contribute to nerve growth factor-mediated neuroprotection.
