Description
The peptide [Lys5,MeLeu9,Nle10]Neurokinin A(4-10) (LMN-NKA) is a selective agonist of the neurokinin NK2 receptor, exhibiting high specificity in binding and functional assays. It demonstrates a 674-fold selectivity for NK2 over NK1 receptors in radioligand binding assays and ≥74-fold selectivity in functional in vitro assays. LMN-NKA induces contractile activity in visceral smooth muscle, particularly in the colon and urinary bladder, across various species including rats, dogs, pigs, and humans.
In experimental models, LMN-NKA rapidly increases bladder and colorectal pressures upon administration, with effects mediated through NK2 receptor activation. The peptide's pharmacological profile includes dose-dependent increases in peak bladder and colorectal pressures, with rapid onset and sustained duration of action. These effects are inhibited by pretreatment with the NK2 receptor antagonist GR 159897, confirming receptor specificity. The peptide's activity is conserved across species, making it a valuable tool for studying NK2 receptor-mediated physiological responses.
