  • Description

    The WIK-14 peptide is a 14-amino acid α-helical antimicrobial peptide (AMP) designed with a sequence of WIKKIWKKIIKKIK, featuring lysine (K), isoleucine (I), and tryptophan (W) residues. Its amphipathic structure enables interactions with bacterial membranes through electrostatic and hydrophobic forces. Substitution of the 12th lysine with glutamic acid (E) yields WIKE-14 (WIKKIWKKIIKEIK), which adopts a helix-to-helix conformation. This structural flexibility enhances its binding capacity to lipopolysaccharides (LPS) via electrostatic interactions with phosphate groups and hydrophobic interactions with lipid A. CD spectroscopy confirms helical formation in LPS and membrane-mimetic environments. Both WIK-14 and WIKE-14 exhibit potent LPS-neutralizing activity, inhibiting pro-inflammatory cytokine release in macrophages by blocking LPS binding to TLR4/MD-2 receptors.
