    TRV056 is a Gq-biased agonist of the angiotensin II type 1 receptor (AT1R), a G protein-coupled receptor (GPCR). Compared to the endogenous agonist angiotensin II (AngII), TRV056 exhibits enhanced Gq coupling and cellular signaling while maintaining similar efficacy towards β-arrestin. Structural studies using double electron-electron resonance (DEER) spectroscopy reveal that TRV056 stabilizes distinct receptor conformations characterized by an outward displacement of transmembrane helix 6 (TM6) and specific rearrangements in intracellular loop 2 (ICL2) and helix 8 (H8). These conformational changes are associated with an accessible transducer-binding site, rationalizing TRV056's enhanced Gq activity. The peptide's C-terminal modifications, particularly at position 8, play a critical role in determining its biased signaling profile by influencing receptor dynamics and transducer coupling.
