Description
The TRV055 peptide is a Gq-biased agonist of the angiotensin II type 1 receptor (AT1R). Compared to the endogenous ligand angiotensin II (AngII), TRV055 exhibits enhanced Gq coupling and cellular signaling while maintaining comparable efficacy towards β-arrestin. Structural studies using double electron-electron resonance (DEER) spectroscopy reveal that TRV055 stabilizes distinct receptor conformations, particularly favoring an "open" conformation (Aopen) with an accessible transducer-binding site. This contrasts with β-arrestin-biased agonists, which stabilize more occluded conformations. The conformational preferences of TRV055 provide insights into the structural basis of biased signaling at AT1R.
