Description
The TRPV1 inhibitory peptide (TIP) is a synthetic nine-amino acid peptide with the sequence Gln-Arg-Ala-Ile-Thr-Ile-Leu-Asp-Thr (QRAITILDT). It is designed to mimic a specific cytosolic region (residues 701–709) of the transient receptor potential vanilloid 1 (TRPV1) channel, which includes the critical phosphorylation site Thr705. TIP functions as a competitive inhibitor by binding to TRPV1 and preventing its phosphorylation and subsequent activation, thereby blocking calcium influx through the channel.
Studies demonstrate that TIP effectively inhibits TRPV1-mediated responses, including ultraviolet (UV)-induced upregulation of matrix metalloproteinases (MMPs) and pro-inflammatory cytokines. The peptide is cell-permeable and retains biological activity in epidermal keratinocytes and skin models, where it attenuates UV-induced molecular and cellular changes without acting as a physical UV filter.

About TFA salt
Trifluoroacetic acid (TFA) is a common counterion from the purification process using High-Performance Liquid Chromatography (HPLC). The presence of TFA can affect the peptide's net weight, appearance, and solubility.
Impact on Net Weight: The TFA salt contributes to the total mass of the product. In most cases, the peptide content constitutes >80% of the total weight, with TFA accounting for the remainder.
Solubility: TFA salts generally enhance the solubility of peptides in aqueous solutions.
In Biological Assays: For most standard in vitro assays, the residual TFA levels do not cause interference. However, for highly sensitive cellular or biochemical studies, please be aware of its presence.
