Description
Recombinant Human TANGO1/MIA3 SH3 Domain Fragment (aa 31-123) - A highly specific research tool designed for studying the molecular mechanisms of collagen VII and lipoprotein transport from the endoplasmic reticulum. This purified fragment contains the complete SH3 functional domain (aa 45-107), enabling targeted investigation of cargo loading processes in large protein secretion pathways.
Transport and Golgi organization protein 1 homolog (TANGO1), encoded by the MIA3 gene, plays a critical role in the specialized transport of cargos too large for conventional COPII-coated vesicles, particularly collagen VII (COL7A1). This endoplasmic reticulum membrane protein functions at ER exit sites by binding to COPII coat subunits Sec23/24 and guiding SH3-bound COL7A1 into growing transport carriers. While essential for collagen VII secretion, TANGO1 also participates in lipoprotein export from the endoplasmic reticulum and is required for proper assembly of COPII components at ER exit sites. The protein localizes specifically to endoplasmic reticulum exit sites (transitional ER) where it remains after cargo loading, rather than being incorporated into COPII carriers, highlighting its role as a stationary cargo receptor in specialized secretion pathways.
