    Teduglutide is a synthetic analog of glucagon-like peptide-2 (GLP-2), a naturally occurring peptide hormone involved in intestinal growth and function. Structurally, teduglutide differs from native GLP-2 by a single amino acid substitution, which confers resistance to degradation by dipeptidyl peptidase IV (DPP-IV), thereby extending its half-life. This modification enhances its biological activity in promoting intestinal mucosal growth. Studies in animal models demonstrate that teduglutide increases villus height, crypt depth, and epithelial cell proliferation in the small intestine and colon. Additionally, it enhances nutrient transport capacity, particularly for glucose and glutamine, across the intestinal epithelium.

    The peptide exerts its effects by binding to the GLP-2 receptor, which is expressed in the gastrointestinal tract. Activation of this receptor signaling pathway stimulates cellular proliferation and inhibits apoptosis, leading to increased mucosal surface area. Teduglutide has been shown to modulate intestinal adaptation following resection, with effects observed in both structural and functional parameters. Its mechanisms include upregulation of absorptive surface area and transient enhancement of nutrient processing capacity in the intestine.
