    The Tat-syn-degron peptide is a synthetic construct designed to reduce intracellular levels of α-synuclein. It comprises three functional domains: a Tat protein transduction domain that facilitates crossing of the blood-brain barrier and plasma membranes, a β-synuclein-derived sequence that binds specifically to monomeric α-synuclein, and a proteasomal targeting degron signal. This design enables the peptide to bind α-synuclein and direct the complex to the proteasome for degradation.

    Experimental evidence demonstrates that the peptide effectively reduces α-synuclein levels in neuronal cultures and in vivo. The reduction is mediated by the proteasomal pathway, as it is prevented by proteasomal inhibitors. The peptide's action is specific to α-synuclein, as it does not affect levels of related synuclein family members or other unrelated neuronal proteins. The knockdown effect is dose-dependent, time-dependent, and transient, peaking approximately 24 hours after administration.
