Description
Suc-AEPF-AMC is a fluorogenic substrate in the assay to measure the peptidyl prolyl isomerase (PPIase) activity of Pin1. AEPF can be utilized by both Pin1 and Par14. The cis to trans isomerization of the Ser - Pro peptide bond proceeds fast, so the assay was performed at 4°C. The reliability of this assay method was confirmed by enzyme kinetic analysis. The identification of Pin1 and Par14 PPIase inhibitors using AEPF as a substrate helped the researchers to investigate the roles of these proteins in cell proliferation and cell cycle progression.
