SMAD3 is a receptor-regulated SMAD (R-SMAD) that serves as a key intracellular signal transducer and transcriptional modulator activated by TGF-beta and activin type 1 receptor kinases. It binds to TRE elements in promoter regions and, upon forming complexes with SMAD4, activates transcription of TGF-beta-regulated genes. SMAD3 also participates in complexes with JUN/FOS at AP-1/SMAD sites to fine-tune transcriptional responses. Functionally, it inhibits wound healing by modulating keratinocyte growth and migration and altering monocyte chemotaxis, while also regulating chondrogenesis, osteogenesis, and bone fracture healing. SMAD3 localizes to both the cytoplasm and nucleus, transitioning to the nucleus upon TGF-beta stimulation when complexed with SMAD4, and its nuclear export is mediated by interactions with RANBP1 following PPM1A phosphatase activity.
