Schlafen family member 11 (SLFN11) is a critical human protein that acts as a guardian of the genome by inhibiting DNA replication and promoting cell death in response to DNA damage. It persistently blocks stressed replication forks by opening chromatin at replication initiation sites, which can unwind DNA and halt fork progression, ultimately leading to cell death. SLFN11 also functions as an antiviral restriction factor with manganese-dependent type II tRNA endoribonuclease activity, specifically impairing the replication of retroviruses like HIV-1, human cytomegalovirus, and some Flaviviruses by exploiting viral codon bias toward A/T nucleotides and inhibiting viral protein synthesis. Additionally, upon DNA damage, it translationally inhibits ATR or ATM without disrupting early DNA damage signaling. The protein is localized to the nucleus and chromosomes, where it is recruited to stressed replication forks and DNA damage sites via interaction with RPA1, underscoring its role in cellular stress responses and antiviral defense mechanisms.
