SGF29 functions as a crucial chromatin reader component within histone acetyltransferase (HAT) SAGA-type complexes, including TFTC-HAT, ATAC, and STAGA complexes. The protein specifically recognizes and binds methylated 'Lys-4' of histone H3, with preferential affinity for the trimethylated form (H3K4me3). Within SAGA-type complexes, SGF29 is essential for recruiting these complexes to H3K4me sites, facilitating targeted histone acetylation. The protein plays a significant role in cellular stress response, particularly during endoplasmic reticulum stress, where it promotes cell survival by directing the SAGA complex to H3K4me sites and enhancing histone H3 acetylation. Additionally, SGF29 demonstrates binding capability to non-histone proteins methylated on lysine residues, such as CGAS monomethylated at 'Lys-506'.
