Description
The peptide KKKVTAQELD is a highly conserved sequence found in staphylococcal enterotoxins (SE), including SEB, SEA, SEE, and TSST-1. Structural analysis reveals that this region encompasses the end of beta sheet 7, beta sheet 8, and the beginning of helix 4, a conformation preserved across SE. Experimental studies demonstrate that synthetic peptides containing this sequence significantly inhibit the transcytosis of SEB across epithelial cell monolayers, reducing movement by up to 85% at a toxin-to-peptide ratio of 1:10. The inhibitory effect extends to other SE, such as SEA, SEE, and TSST-1, indicating a conserved functional role in toxin translocation. Antisera generated against this peptide recognize native SEB and also block transcytosis, further confirming its involvement in epithelial barrier crossing.
