Sperm Acrosome Membrane-Associated Protein 3 (SPACA3), also known as Cancer/Testis Antigen 54, is a sperm surface membrane protein implicated in sperm-egg plasma membrane adhesion and fusion during fertilization. The protein may function as a receptor for N-acetylglucosamine oligosaccharide residues present in the egg extracellular matrix. SPACA3 localizes to the acrosome membrane in cytoplasmic vesicles and exists as both membrane-anchored (isoform 1) and secreted (isoform 2) forms. While the processed form shows no detectable bacteriolytic activity, the full-length protein plays a crucial role in reproductive processes. The recombinant fragment (aa 85-215) contains the complete C-type lysozyme domains, enabling focused investigation of these functional regions without interference from other protein segments.
