Description
S-2238 is a synthetic chromogenic substrate specifically designed for thrombin assays. It consists of the peptide sequence H-D-Phenylalanyl-L-Pipecolyl-Arginine-p-nitroaniline (pNA), which mimics the N-terminal portion of the fibrinogen A alpha chain, the natural substrate of thrombin. Thrombin cleaves the terminal arginine-pNA bond, releasing the chromophore p-nitroaniline, enabling spectrophotometric quantification at 405 nm. The inclusion of phenylalanine and pipecolic acid enhances thrombin specificity and hydrolysis efficiency.
The substrate demonstrates high solubility and sensitivity to thrombin, allowing shorter reaction times and reduced plasma interference from factors like hyperbilirubinemia or hyperlipidemia. In assays, S-2238 is combined with polybrene to neutralize heparin and optimize thrombin activity measurement. Its stability and reproducibility make it a robust tool for studying serine proteases and their inhibitors in coagulation systems.
