Description
Ro 25-1553 is a synthetic 31-amino acid analogue of vasoactive intestinal peptide (VIP). It is characterized by high selectivity for the VPAC2 receptor subclass. In vitro studies on guinea-pig isolated trachea demonstrate that Ro 25-1553 induces a concentration-dependent and complete relaxation of precontracted tracheal smooth muscle. Its efficacy is comparable to that of the long-acting β2-adrenoceptor agonist formoterol, though it is 3 to 7 times less potent on a molar basis. The compound exhibits a rapid onset of action and an additive interaction with formoterol. Furthermore, Ro 25-1553 concentration-dependently inhibits nerve-induced contractions in vagus nerve-trachea preparations and retains activity when administered intraluminally, albeit at higher concentrations.
