In response to nutrients, growth factors, or amino acids, Rheb activates MTOR's kinase activity, enhancing phosphorylation of downstream targets like RPS6KB1 and EIF4EBP1. It localizes to lysosomal, Golgi, endoplasmic reticulum, and endomembrane systems via lipid anchoring, with farnesylation required for lysosomal membrane recruitment and mTORC1 activation. Under starvation conditions, Rheb is inactivated by the TSC-TBC complex, highlighting its role in metabolic regulation and cellular homeostasis.
