Description
PRO20 is a 20-amino-acid peptide antagonist of the (pro)renin receptor (PRR). Its sequence corresponds to the first 20 amino acids of the (pro)renin prosegment. This peptide exhibits high specificity for both human and mouse PRR, functioning by competitively inhibiting the binding of (pro)renin to its receptor. In experimental models, subcutaneous administration of PRO20 has been shown to modulate hepatic lipid metabolism. Mechanistically, treatment with PRO20 is associated with the attenuation of diet-induced increases in the expression of key lipid synthesis regulators in the liver, including the transcription factor PPARγ and the enzyme glycerol-3-phosphate acyltransferase 3 (GPAT3). These molecular effects correlate with a reduction in hepatic triglyceride accumulation and lipid droplet formation.
