Description
Recent research has shown that a specific protein called CypA plays a key role in the aggregation and toxicity of α-synuclein protein in the body. This protein catalyses the isomerisation of proline 128 in the C-terminal structural domain of α-synuclein, which contributes to its pathogenicity. Additionally, the hydrophobic sequence 47GVVHGVATVA56 forms a second CypA-binding site, which is crucial for the age-dependent aggregation and toxicity of αSyn in the nematode model of Hidradenitis elegans. By deleting this sequence, it is possible to prevent αSyn aggregation and toxicity. Furthermore, this sequence also plays a role in αSyn-mediated vesicle fusion by altering the conformational properties of the protein upon lipid binding. This region could be targeted to prevent αSyn aggregation in disease.
