Protection of telomeres homolog 1 (POT-1), encoded by the pot-1 gene in Caenororhabditis elegans, is a telomeric DNA-binding protein that specifically recognizes single-stranded C-rich repeat sequences, particularly the 5'-GCCTAA-3' motif. It plays a critical role in safeguarding the 5' end of the C-rich telomeric strand and functions redundantly with POT-2 to negatively regulate telomerase-mediated telomere extension. Additionally, POT-1 is involved in the telomerase-independent alternative lengthening of telomeres (ALT) pathway and, through interaction with SUN-1, anchors telomeres to the nuclear envelope in embryos. The protein localizes to the nucleus, nuclear envelope, and chromosome telomeres, with higher expression in meiotic pachytene nuclei and presence in mitotic nuclei, underscoring its importance in telomere maintenance and genomic stability.
