Protein O-glucosyltransferase 3 (POGLUT3) is an endoplasmic reticulum-resident enzyme that catalyzes the transfer of glucose from UDP-glucose to serine residues within the consensus sequence C-X-N-T-X-G-S-F-X-C in extracellular EGF repeats. It also exhibits less efficient xylosyltransferase activity using UDP-xylose. POGLUT3 specifically targets EGF domains of proteins including NOTCH1, NOTCH3, FBN1, FBN2, and LTBP1, playing a crucial role in regulating the transport of NOTCH receptors to the plasma membrane and thereby modulating Notch signaling pathways involved in cellular differentiation and development.
