Description
A short, 12-meric antimicrobial peptide, Pap12-6, was designed by substituting amino acids from the N-terminal 12 residues of papiliocin (Pap12-1) to alter cationicity and amphipathicity. Pap12-6 exhibits broad-spectrum antibacterial activity, particularly against multidrug-resistant Gram-negative bacteria. It kills bacteria by permeabilizing the bacterial membrane, as demonstrated by dye leakage, membrane depolarization, and electron microscopy. Pap12-6 also reduces secretion of NO, TNF-α, and IL-6, showing anti-inflammatory activity via a TLR4-mediated NF-κB signaling pathway.
