    P1 (FLYTLLREV) is a 9-mer HLA-A02:01-restricted cytotoxic T lymphocyte (CTL) epitope derived from the Six-Transmembrane Epithelial Antigen of the Prostate 1 (STEAP1), mapping to the 86–94 amino acid region of the STEAP1 protein (UniProt ID: Q9UHE8). This peptide exhibits a high predicted binding affinity to HLA-A02:01 (percentile rank 0.03, binding score 0.94) and robust structural compatibility with the MHC class I binding groove, as validated by molecular docking with a binding energy of −246.57. Characterized by an α-helical secondary structure in aqueous physiological conditions, FLYTLLREV elicits potent antigen-specific IFN-γ secretion and efficient activation of polyfunctional CD8+ T cells with co-expression of IFN-γ and TNF-α, demonstrating strong immunogenicity in HLA-A*02:01 transgenic murine models. It also shows high sequence conservation across homologs and favorable proteasomal processing potential, supporting its natural presentation via the endogenous MHC class I antigen presentation pathway.
