    LP17 is a synthetic peptide derived from the complementary determining region-3 and the "F" β-strand of the extracellular domain of TREM-1. It functions as an inhibitory peptide by competitively binding to TREM-1, thereby blocking its interaction with natural ligands. LP17 has been shown to effectively suppress TREM-1-mediated signaling pathways, including the activation of SYK and downstream pro-inflammatory cascades such as CARD9/NF-κB and NLRP3/caspase-1. This peptide also attenuates microglial M1 polarization and reduces the production of inflammatory cytokines and chemokines.

    In experimental models, LP17 demonstrates the ability to mitigate neuroinflammatory responses by inhibiting TREM-1/SYK interactions. This leads to decreased levels of inflammatory mediators such as IL-1β, IL-18, and IL-6, as well as reduced chemokine expression. Additionally, LP17 has been observed to diminish microglial pyroptosis by lowering the levels of GSDMD and its N-terminal fragment, which are critical for the formation of membrane pores and the release of intracellular inflammatory factors. The peptide's mechanism highlights its potential role in modulating innate immune responses in the central nervous system.
