Description
KFP-1 is a 17-residue peptide (TEVPAINTIASAEPTVH) isolated from kefir, a fermented dairy beverage. This peptide is characterized as a calcium-binding peptide that facilitates intestinal calcium absorption through TRPV6 channels. In vitro studies demonstrate that KFP-1 promotes osteoblast differentiation and mineralization. It upregulates key osteogenic markers and transcription factors, including ALP, Col1a1, OCN, RUNX2, OSX, BMP-2, and β-catenin.
Concurrently, KFP-1 exhibits inhibitory effects on osteoclast differentiation and bone resorption. It downregulates osteoclastic markers such as cathepsin K, TRAP, and NFATc1, and suppresses RANKL-induced signaling pathways, including p38 MAPK, JNK, ERK, and NF-κB. Biodistribution analyses indicate that KFP-1 can reach bone tissue following oral administration. The collective data indicate that KFP-1 possesses a dual function in modulating bone remodeling by enhancing osteogenic activity and inhibiting osteoclastic activity.
