    Lysine-specific demethylase 7A (KDM7A) is a nuclear histone demethylase crucial for brain development, functioning as a key regulator of the histone code. It specifically demethylates dimethylated lysine residues at positions 9, 27, and 36 (H3K9me2, H3K27me2, H3K36me2) on histone H3, as well as monomethylated lysine 20 on histone H4 (H4K20Me1). The enzyme's specificity is modulated by histone context: it demonstrates high activity toward H3K27me2 in the presence of H3K4me3 binding, while preferentially targeting H3K9me2 in its absence. Notably, its activity toward H4K20Me1 requires nucleosomal substrates rather than histone octamers, highlighting its biological relevance in chromatin remodeling processes.
