    Integral Membrane Protein 2B (ITM2B) is a human protein that plays crucial regulatory roles in neuronal function and amyloid pathology. The full-length protein functions as a key modulator of amyloid-beta A4 precursor protein (APP) processing, acting as an inhibitor of amyloid-beta peptide aggregation and fibril deposition. ITM2B demonstrates multiple functional forms through proteolytic processing: immature BRI2 (imBRI2) is cleaved in the Golgi apparatus to produce membrane-bound mature BRI2 (mBRI2) and the secreted Bri23 peptide. Mature BRI2 strongly reduces secretion of amyloid-beta proteins 40 and 42, while the Bri23 peptide specifically prevents aggregation of APP amyloid-beta protein 42 into toxic oligomers. The protein also contributes to neurite outgrowth induction and functions as a protease inhibitor by blocking secretase access to APP cleavage sites. Localized primarily to Golgi and plasma membranes as a single-pass type II membrane protein, ITM2B's various processed forms occupy distinct subcellular compartments, with mature BRI2 requiring transport to the cell membrane for APP inhibition activity and Bri23 peptide being secreted into cerebral spinal fluid.
