Recombinant Human IRGM (Immunity-related GTPase family M protein) Fragment (aa 1-181) - A high-purity protein fragment encompassing the complete IRG-type G domain, essential for studying innate immunity, autophagy regulation, and inflammatory response mechanisms in human cellular systems.
IRGM is a crucial immunity-related GTPase that plays pivotal roles in innate immunity and inflammatory response regulation. This dynamin-like protein binds to intracellular membranes and promotes membrane remodeling and trafficking. It is essential for clearance of acute protozoan and bacterial infections by facilitating phagosome-lysosome fusion for efficient microbial degradation. IRGM regulates selective autophagy pathways, including xenophagy and mitophagy, through multiple mechanisms: activating AMPK to phosphorylate ULK1 and BECN1, promoting ULK1-BECN1 coassembly, and influencing BECN1 complex composition. The protein also modulates lysosomal biogenesis by promoting TFEB dephosphorylation and assists with autophagosome formation. Importantly, IRGM serves as a key negative regulator of inflammatory and interferon responses by promoting mitophagy and mediating autophagy-dependent degradation of inflammatory effectors. It localizes to multiple intracellular membranes including Golgi apparatus, phagosomes, autophagosomes, lysosomes, late endosomes, and mitochondria, behaving as an integral membrane protein that remains associated with maturing phagosomes in a nucleotide-binding dependent manner.
