    The IA9 peptide is a rationally designed 9-mer sequence (IFLIKILAA) derived from human TREM-2182–190, developed using the signaling chain homooligomerization (SCHOOL) model of cell signaling. This peptide employs a ligand-independent mechanism to inhibit TREM-2 by blocking its interaction with the signaling partner DAP-12 in the cell membrane. IA9 can be delivered either as a free peptide or incorporated into lipopeptide complexes (LPC) for targeted intracellular delivery to macrophages. In experimental collagen-induced arthritis (CIA), IA9 significantly suppresses joint inflammation and damage, reduces proinflammatory cytokine release, and decreases synovial tissue expression of inflammatory markers, demonstrating its potential as a modulator of TREM-2 signaling in inflammatory conditions.
