    Herpes Virus Inhibitor 1 (HVI-1) is a synthetic nonapeptide with the sequence YAGAVVNDL, corresponding to the carboxyl-terminal nine amino acids of the small subunit of herpes simplex virus type 1 ribonucleotide reductase. This peptide specifically inhibits the viral enzyme by competing with the R2 subunit for binding to the R1 subunit, thereby disrupting the formation of the active R1/R2 dimer complex. The inhibitory activity is selective for herpesvirus ribonucleotide reductases, as their R2 carboxyl termini share homology that differs substantially from reductases of other species.

    To facilitate intracellular delivery, the nonapeptide has been genetically linked to the B subunit of Escherichia coli heat-labile enterotoxin (EtxB), which binds to GM1 ganglioside receptors on eukaryotic cell surfaces and enters cells via receptor-mediated uptake. The resulting fusion protein retains the ability to assemble into a stable pentamer, binds specifically to the HSV-1 R1 subunit, and this interaction can be competitively disrupted by the free nonapeptide. This carrier-mediated approach enables the evaluation of the peptide's intracellular activity against viral ribonucleotide reductase in cell culture systems.
