    GsMTx4 is a 34-amino acid peptide toxin isolated from the venom of the Chilean rose tarantula spider Grammostola spatulata. It belongs to the inhibitor cystine knot (ICK) family, characterized by three disulfide bonds forming a stable structural motif. GsMTx4 specifically inhibits stretch-activated cation channels, though its mechanism involves interaction with the lipid bilayer surrounding the channels rather than direct binding to channel proteins.

    Structural analysis reveals that GsMTx4 folds into two interconnected rings, a conformation stabilized by its cysteine knots. This peptide has been shown to block mechanosensitive currents in various cell types, including astrocytes and cardiomyocytes, suggesting a broad modulatory role in mechanotransduction pathways. Its unique selectivity makes it a valuable tool for studying mechanosensitive ion channels and their physiological functions.
