Description
This recombinant protein product features a stable and soluble fragment (amino acids 65-237) of the Eukaryotic translation initiation factor 2-alpha kinase 1 (EIF2AK1) from Homo sapiens. Eukaryotic translation initiation factor 2-alpha kinase 1 (EIF2AK1), also known as Heme-controlled repressor (HCR) or HRI, is a key sensor and activator of the integrated stress response (ISR) in humans. It phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (EIF2S1/eIF-2-alpha) under various stress conditions, including heme deficiency, oxidative stress, osmotic shock, mitochondrial dysfunction, and heat shock. This phosphorylation globally attenuates cap-dependent protein synthesis while promoting the translation of stress-specific mRNAs, such as ATF4, enabling cellular reprogramming and adaptation. EIF2AK1 uniquely senses heme concentrations through moderate-affinity binding, inhibiting its kinase activity under normal conditions and activating it during heme depletion. It plays essential roles in hemoglobin synthesis in red blood cells, protection during iron deficiency anemias, and mitophagy activation in response to mitochondrial damage via interactions with DELE1. The protein fragment (65-237) was computationally selected to provide a stable, soluble unit for detailed structural and functional analyses.
