    DP7 is a synthetic antimicrobial peptide (AMP) derived from the parent peptide HH2 through an amino acid-based activity prediction method. This 12-mer peptide, with the sequence VQWRIRVAVIRK, exhibits broad-spectrum antimicrobial activity against both Gram-positive and Gram-negative bacterial strains. Compared to HH2, DP7 demonstrates enhanced antimicrobial efficacy while maintaining lower cytotoxicity, as evidenced by hemolysis and cell viability assays.

    Electron microscopy studies reveal that DP7 disrupts the outer membrane of Staphylococcus aureus without causing cytoplasmic leakage, distinguishing its mechanism from typical pore-forming AMPs. The peptide's amphipathic and cationic properties facilitate selective binding to bacterial membranes, leading to structural damage and inhibition of bacterial growth. DP7 represents a rationally designed AMP with optimized activity and reduced host cell toxicity.
