    Diazepam binding inhibitor-34 (DBI-34), also known as TTN, is a 34-amino acid neuroactive peptide derived from the proteolytic cleavage of acyl-CoA-binding protein (ACBP). ACBP, a highly conserved protein with a primary intracellular role in fatty acid metabolism, is secreted from astrocytes via an unconventional, brefeldin A-insensitive pathway that involves components of the autophagy machinery. Following secretion, ACBP is rapidly processed extracellularly by a cell surface-associated protease activity to generate the bioactive DBI-34 peptide.

    This peptide is classified as an endozepine and functions as an endogenous ligand for the GABAA receptor. Its production in astrocytes can be rapidly induced within minutes by specific neurosteroids, such as pregnenolone sulfate and cortisol, as well as by cellular depolarization. The secretion and subsequent processing of ACBP into DBI-34 represent a conserved mechanism for intercellular signaling, highlighting a non-classical pathway for peptide generation and release in the central nervous system.
