    The DAPK substrate peptide, KKRPQRRYSNVF, was identified through a combination of structural analysis and positional scanning substrate library synthesis. This peptide serves as an efficient substrate for Death-associated protein kinase (DAPK), a calmodulin-regulated serine/threonine kinase implicated in apoptosis and tumor suppression. The peptide features a core phosphorylation motif, RXXSX, with key residues at positions P-3 (arginine) and P0 (serine). Structural studies of the DAPK catalytic domain revealed unique features, such as a basic loop and specific amino acid interactions, that contribute to substrate recognition. The peptide's Km value of approximately 9 μM makes it suitable for enzymatic assays and further investigations into DAPK's catalytic properties.
