Description

Disabled homolog 2 (DAB2) is an adapter protein that acts as a clathrin-associated sorting protein (CLASP), essential for clathrin-mediated endocytosis of cargo proteins like the LDL receptor, integrin beta-1, and CFTR. It binds clathrin and phosphatidylinositol 4,5-bisphosphate, facilitating the recruitment of cargo to clathrin-coated pits, often independently of the AP-2 complex. DAB2 is involved in key signaling pathways, including TGF-beta receptor signaling, where it promotes SMAD2 phosphorylation, and Wnt/beta-catenin signaling, where it may inhibit canonical pathways by stabilizing the beta-catenin destruction complex. It also plays roles in cellular differentiation, epithelial to mesenchymal transition during embryogenesis, and myeloid cell differentiation, and it may function as a tumor suppressor. Subcellularly, DAB2 localizes to the cytoplasm, clathrin-coated vesicles, and clathrin-coated pits, with dynamic redistribution during mitosis.
