    [D-Leu2]deltorphin is a 17-amino acid opioid peptide isolated from methanol extracts of the skin of the Brazilian hylid frog Phyllomedusa burmeisteri. Its sequence is Tyr-D-Leu-Phe-Ala-Asp-Val-Ala-Ser-Thr-Ile-Gly-Asp-Phe-Phe-His-Ser-Ile-NH2, featuring a D-leucine residue at position 2. This peptide belongs to the deltorphin subfamily, characterized by the presence of a D-amino acid in the second position, which is crucial for bioactivity in related peptides.

    The natural [D-Leu2]deltorphin exhibits low affinity for δ-opioid binding sites in both peripheral tissues and the central nervous system. However, a shorter synthetic amidated analogue, [D-Leu2]deltorphin(1-10), demonstrates significant agonistic potency and selectivity for δ-opioid receptors, comparable to enkephalins. Another truncated analogue, [D-Leu2]deltorphin(1-7), shows minimal activity peripherally but binds to both δ and μ opioid sites in rat brain membranes. This peptide is part of a group of natural animal-derived peptides containing D-amino acids, all found at position 2 of their sequences.
