Description
C-II-a, also known as ApoC-II-a, is a novel bihelical amphipathic peptide designed to mimic the properties of apolipoprotein C-II. It consists of two distinct helical domains: an N-terminal 18A amphipathic helix that confers high affinity for lipid binding to lipoproteins, and a C-terminal helix derived from the last helix of apolipoprotein C-II, which contains the critical motif for activating lipoprotein lipase (LPL). This structural design enables the peptide to effectively associate with lipoprotein surfaces and stimulate LPL-mediated lipolysis of triglycerides.
In vitro characterization demonstrates that the C-II-a peptide is a potent activator of LPL, exhibiting efficacy comparable to the full-length apolipoprotein C-II protein in promoting lipolysis using both artificial lipid emulsions and native lipoprotein substrates. The peptide also facilitates cholesterol efflux via the ATP-binding cassette transporter ABCA1. Biophysical analyses, including structural modeling and circular dichroism spectroscopy, confirm its amphipathic helical conformation and interaction with lipid surfaces.
