Description
The 8DSS peptide is a synthetic oligopeptide consisting of eight tandem repeats of the aspartate-serine-serine (DSS) amino acid sequence. This sequence is the fundamental repetitive unit found within dentin phosphoprotein (DPP), a non-collagenous protein that plays a critical role in the biomineralization process of dentin. The peptide is characterized by its molecular weight of approximately 2331.4 Da and contains multiple negatively charged amino acid residues.
In vitro studies demonstrate that the 8DSS peptide exhibits a strong binding affinity to demineralized type I collagen matrices, primarily through electrostatic interactions. When applied to such substrates in a mineralizing solution, the peptide functions as a nucleation template, promoting the precipitation and growth of hydroxyapatite nanocrystals. These newly formed minerals deposit on the collagen fibril surfaces and within dentinal tubules, leading to microstructural restoration and a significant improvement in the nano-mechanical properties, such as hardness and elastic modulus, of the treated substrate.
