Anti-succinyllysine monoclonal antibody detects proteins post-translationally modified by lysine succinylation. The antibody recognizes succinyllysine in a wide range of sequence contexts, both in histones and non-histone substrates.
The pan anti-succinyllysine monoclonal antibody is developed by using succinylated-KLH conjugates as immunogen. The product specifically recognizes proteins with lysine succinylated residues but not the structurally similar modified lysine residues or non-modified lysine residues. Besides common applications in Western blotting, the antibody has been well utilized for proteomic screening to lysine succinylated substrates from protease-digested proteins both in prokaryotic and eukaryotic cells.
Antibody is supplied in PBS with 50% glycerol and 0.01% sodium azide.
Store product at -20 ℃. Avoid repeated freeze/thaw cycles. Stable for 12 months from date of receipt.
For Western blotting, incubate membrane with diluted antibody in 5% nonfat milk, 1 x TBS, 0.1% Tween-20 at 4 degree centigrade; with gentle shaking, overnight.
Use at an assay dependent concentration. Optimal dilutions/concentrations should be determined by the end user.

Dot blotting analysis on 20 ng of lysine succinylated peptide library (lane 1), lysine succinylated BSA (lane 2), selected histone lysine succinylated peptides (lane 3 to lane 8), lysine acetylated BSA (lane 9), lysine butyrylated BSA (lane 10), lysine propionylated BSA (lane 11), lysine crotonylated BSA (lane 12), unmodified lysine peptide library (13), unmodified BSA (14) using anti-succinyllysine mAb. B. Western blotting analysis on 30ug of HeLa, C2C12 and COS crude proteins using anti-succinyllysine mAb.
Lysine succinylation is a recently identified novel Protein Post-translational Modification. Lysine succinylation is evolutionarily conserved and abundantly present in proteins in both eukaryotic and eukaryotic cells, suggesting its roles in regulating cellular physiology. This antibody will facilitate studies of lysine succinylation biology by identifying its substrates and studying its function.
