NZCYM Broth Preparation and Recipe

NZCYM is a microbial growth medium used for the cultivation of E. coli and lambda bacteriophage. This nutrient-rich microbial broth contains peptides, amino acids, and water-soluble vitamins in a low-salt formulation. NZCYM Broth has been used for non-selective cultivation of lambda, MS2, T7, CLP-P3, Phi29, and Goe1 bacteriophages, and E. coli strains for cloning, DNA plasmid production, and production of recombinant proteins. It is also suitable for selective cultivation when appropriate antibiotics are added.

  1. Prepare 0.8 L of H2O in a suitable container.
  2. Add 10 g of NZ Amine A to the solution.
  3. Add 0.976 g of Magnesium sulfate anhydrous to the solution.
  4. Add 5 g of Sodium chloride to the solution.
  5. Add 5 g of yeast extract to the solution.
  6. Add 1 g of Casamino Acids to the solution.
  7. Add H2O until the volume is 1 L.
  8. Adjust pH with NaOH sol. to 7.0. Autoclave for 15 min on liquid cycle.
