- Prepare 0.8 L of H2O in a suitable container.
- Add 2.5 g of tryptone to the solution.
- Add 1.25 g of yeast extract to the solution.
- Add 0.1 L of glycerol to the solution.
- Add H2O until the volume is 1 L.
- Sterilize the medium by passing it through a prerinsed 0.22-µm filter. Store in 2.5-mL aliquots at 4°C.
